To enter his plea, the 80-year-old showed up before the federal court in Central Islip, Long Island, New York.
He is released on a $10 million (£7.7 million) bond and is scheduled to return to court on December 12.
Using the promise of modeling for the clothes store, Jeffries, his lover Matthew Smith, and a third man, James Jacobson, are accused of luring men into attending sex parties for years.
Before today’s session, Jacobson, 71, a Jeffries employee, stated that he intended to enter a not guilty plea.
After prosecutors expressed concerns that the dual American and British citizen would leave the country, Smith also showed up in court and was placed under arrest.
Charges made public earlier this week allege that 15 accusers were forced to attend drug-fueled sex parties by “force, fraud, and coercion” in which the men were sometimes told to wear costumes, take alcohol and muscle relaxants, use sex toys, and endure painful penile injections that resulted in erections.
