Left-arm spinner Zafar Gohar claimed his fifth wicket as Sussex trailed 144 to win in 49 overs, but the unflappable Cheteshwar Pujara led Sussex to victory with an unbeaten 44.
With a seventh-wicket stand of 87, Miles Hammond (77) and Gohar (52) gave Gloucestershire optimism as they batted through the morning session to record their second half-centuries of the game.
Gloucestershire was dismissed for 205 as Hammond fell victim to off-spinner Jack Carson twice in three balls. Jayden Seales then ended the innings with wickets from consecutive deliveries.
